   MECHELEN (AFN) - Biotechnologiebedrijf Galapagos begint een tweede fase I-studie met een kandidaatmedicijn tegen gewichtsverlies en verlies van spiermassa (cachexia) en mogelijke andere indicaties zoals de ziekte van Duchenne.
   
   De studie volgt op de eerste studie met het kandidaatmedicijn, GLPG0492, waarin volgens Galapagos goede veiligheids- en farmacologische eigenschappen zijn aangetoond bij eenmaaldaagse orale dosering. ,,Het doel van deze studie is een relevante indicator van de werking van GLPG0492 op de spierfunctie bij gezonde vrijwilligers aan te tonen'', aldus de onderneming woensdag.

   Reversal boosts ATryn prospects
   European panel now recommends GTC anti-clotting drug’s surgical use

   FRAMINGHAM — A European drug regulation committee reversed itself yesterday and recommended that a GTC Biotherapeutics Inc. anti-clotting drug drawn from the milk of genetically altered goats be approved for use in surgical patients.
   
   The move clears away another hurdle for ATryn, a long labored-over drug that could become the world’s first human medicine produced in genetically engineered, or transgenic, animals. It also boosts GTC, which saw its stock tumble in February after European regulators said the company’s studies were insufficient to approve the drug.
   
   “Certainly, for the company as a whole, this is a tremendous validation,” said Thomas E. Newberry, vice president of corporate communications.
   
   GTC stock, which trades under the symbol GTCB, nearly doubled in value yesterday, rising 83 cents, to $1.81, on the Nasdaq Stock Market.
   
   ATryn’s key ingredient is recombinant human antithrombin, a blood protein that blocks one of the body’s clotting substances. GTC has developed goats that carry the human gene for antithrombin. Female goats with the gene produce the protein in their milk, and the protein can be extracted and purified for use in human treatments.
   
   The company operates a farm in Charlton for genetically altered, or transgenic, goats.
   
   Final approval to sell ATryn could come from the European Commission in about three months, according to GTC. The initial European market for ATryn would be surgical patients with an inherited antithrombin deficiency, a relatively small population that regulators estimate could total one in every 3,000 to 5,000 patients.
   
   Yet ATryn could be a foot in the door for GTC as it seeks to widen the use of ATryn and the acceptance of transgenic medicines. GTC is working with LEO Pharma A/S to develop ATryn in Europe as a treatment for clotting in sepsis patients who have acquired an antithrombin deficiency, a market with an estimated 220,000 Europeans a year.
   
   GTC is also conducting U.S. studies of ATryn in an effort to seek Food and Drug Administration approval in mid-2007.
   
   Rodman & Renshaw analyst Navdeep S. Jaikaria, who yesterday raised his rating on GTC stock to “outperform,” told clients in a note that ATryn approval was “in fact a milestone achievement for GTCB. This is the first time ever that a transgenic goat product is approved for any indication, which serves to validate GTCB’s proprietary technology platform.”
   
   In a written release issued yesterday in London, the European Medicines Agency Committee for Medicinal Products for Human Use said it had initially considered GTC’s patient studies too small. The studies contained information on five surgical patients and nine women in childbirth. At first, the committee disregarded the data from the pregnant women and told GTC it had not provided data on at least 12 patients, as required.
   
   The European committee also questioned manufacturing changes that GTC made after the data had been collected.
   
   GTC asked the committee to re-examine the data, and the committee convened a panel of European experts on blood and blood clotting. Yesterday, the committee reported that it concluded it could use the data from all the patients because all faced the risk of blood clotting, but it restricted the drug’s use to nonpregnant patients.
   
   The committee also reported that manufacturing changes made by GTC were slight and that “the benefits of ATryn outweigh its risks.”
   
   Regulators have been cautious about the use of animals to produce medicines, even as many researchers have devoted their careers to the field and companies have labored on limited funding to advance the technology, said Eric W. Overstrom, professor and head of the department of biology and biotechnology at Worcester Polytechnic Institute.
   
   “The whole concept of a bioreactor, an animal-based bioreactor, this will really push that forward now,” Mr. Overstrom said. “If they’re (GTC) the first ones in, then I think you’re going to see a rollout, a domino effect.”
   
   News of GTC’s advance raised the price of at least one other transgenics company. Shares of Pharming Group NV of the Netherlands rose 15 percent, to 3.71 euros, in European trading.
   
   GTC has long collaborated with corporate partners interested in exploring new production methods, such as Abbott Laboratories and Bristol-Myers Squibb. The company has also worked, with government backing, on a malaria vaccine that could be produced in goat milk.
   
   An approval in Europe, however, could bolster GTC’s ability to raise money and attract partners. The company ended the first quarter of this year on April 2 with a net loss of $8.5 million, or 14 cents a share, and a cash cushion of $15.6 million.
   
   Mr. Newberry of GTC said the European recommendation allows the company to look at a variety of new options.
   
   “Clearly, with this sort of validation, ‘Great science, but can you do anything with it?’ is a long way to being answered,” he said.

   Als die hard Pharmer en bijna 10 jaar lid ben je natuurlijk op de hoogte van de verwantschap van Pharming met GTC.
   Wanneer je ook nu nog blijft suggereren dat dit vele jaren oude jubelbericht een nieuwtje is,wordt het pas echt bedenkelijk. Lees het ook eerst eens door alvorens het hier te plaatsen.

    Clopidogrel tablets USP are indicated for the treatment for acute coronary syndrome, recent myocardial infarction (MI), recent stroke, or established peripheral arterial disease.
    
    Sun Pharma's clopidogrel tablets USP, 75 mg are generic equivalent to Plavix tablets of Sanofi-aventis.
    
    Plavix is used to prevent blood clots after a recent heart attack or stroke, and in people with certain disorders of the heart or blood vessels.

    Dyax Announces Treatment of First Patient in Phase 2 Trial of Ecallantide for Acute Treatment of ACE Inhibitor-Induced Angioedema
    
    CAMBRIDGE, Mass.--(BUSINESS WIRE)-- Dyax Corp. (NASDAQ: DYAX) announced today treatment of the first patient in a Phase 2 trial evaluating the efficacy and safety of ecallantide, a potent, selective, reversible inhibitor of plasma kallikrein, for acute treatment of angiotensin converting enzyme (ACE) inhibitor-induced angioedema. This form of angioedema, or rapid swelling of the dermis, mucosa or other tissues, most commonly occurs in the head, face and tongue, and can be severe or life threatening. Ecallantide is approved under the brand name KALBITOR® in the United States for the treatment of acute attacks of hereditary angioedema (HAE) in patients 16 years of age and older.
    
    "ACE inhibitor-induced angioedema is a serious, potentially life threatening inflammatory condition, for which there are no approved treatments," said Howard Klausner, M.D., associate research director, Department of Emergency Medicine, at the Henry Ford Hospital in Detroit, MI, one of multiple sites involved in the study. "Although this condition is typically treated with standard allergy drugs, these medications are ineffective. Ecallantide could offer benefits in the ACE inhibitor-induced angioedema setting."
    
    The Phase 2, double-blind, placebo-controlled, randomized trial is expected to compare the safety and effectiveness of conventional therapy treatment plus ecallantide to conventional therapy treatment plus placebo in 176 patients enrolled at 40 U.S. clinical sites. The trial includes four treatment arms (N=44 per arm), three active comparator arms investigating 10 mg, 30 mg and 60 mg subcutaneous doses of ecallantide, and a placebo comparator arm. The primary endpoint is patient discharge from the emergency room by 6 hours post dosing.
    
    "We believe that ecallantide's application extends beyond HAE into multiple inflammatory disorders, and our ACE inhibitor-induced angioedema Phase 2 study marks an important step toward realizing this potential," commented Gustav Christensen, President and Chief Executive Officer of Dyax.     di 13 sep 2011, 09:14 Miljoenen voor ontwikkeling medicijn LEIDEN (AFN) - De Leidse biofarmaceutische onderneming Prosensa krijgt 31 miljoen euro van GlaxoSmithKline (GSK) voor de verdere ontwikkeling van een medicijn tegen de ziekte van Duchenne, een tot nu onbehandelbare erfelijke spierziekte bij jongetjes. Dit heeft de onderneming dinsdag bekendgemaakt.
    
    In samenwerking met GlaxoSmithKline gaat Prosensa drie kandidaat-medicijnen tegen de spierziekte in de volgende ontwikkelingsfase brengen. Het programma is een onderdeel van de bestaande samenwerkingsovereenkomst uit 2009 ter waarde van een half miljard euro.
    
    De kandidaat-medicijnen zullen naar verwachting in de eerste helft van 2012 de klinische fase bereiken. Prosensa en GSK gaan daarnaast ook een onderzoek doen naar het natuurlijke beloop van de ziekte van Duchenne.

    Prosensa is in 2004 opgericht door Herman de Boer, de Leidse professor die ondermeer Pharming oprichtte en zijn naam leende aan de befaamde Stier Herman. Inmiddels werken ruim 65 werknemers bij het bedrijf.

    Nederlands farmabedrijf haalt € 29 mln op
    
    Het biofarmaceutische bedrijf AM-Pharma uit Bunnik heeft ruim € 29 mln opgehaald bij beleggers en farmabedrijven voor de ontwikkeling van een medicijn tegen acuut nierfalen.
    
    De investeringsronde is begeleid door Ysios Capital partners en Kurma Life science partners, ondersteund door een consortium met onder meer de farmaconcerns Abbott en Shire. Bestaande investeerders zijn Forbion Capital Partners en Inventages Venture Capital.
    
    Het bedrijf had het middel met een koeien-enzym als actief bestanddeel reeds met succes getest. De Europese autoriteit EMA verlangt echter een menselijk eiwit als actief ingrediënt. AM-Pharma heeft die humane stof inmiddels op kleine schaal geproduceerd en gaat die nu met het opgehaalde kapitaal testen op een groep van 240 patiënten.
    
    Erik van den Berg, ceo van Am-Pharma, ziet de succesvolle financiering als een bewijs van vertrouwen bij de beleggers. ‘Dat blijkt ook door het toetreden van farmabedrijven. De beleggers zien bovendien dat er een beperkt risico is, aangezien we het onderzoek al een keer hebben doorlopen.’
    
    Acuut nierfalen treedt in de wereld jaarlijks op bij 2 miljoen patiënten bij infecties, na een zware operatie, of door bijwerkingen van antibiotica en anti-kankermiddelen. Jaarlijks kost dat 700.000 mensen het leven. De markt heeft een omvang van circa $2 mrd.
    
    Van den Berg schat de duur van de nieuwe onderzoeksfase op circa vier jaar. ‘Daarna gaan we proberen het medicijn onder te brengen bij een farmaconcern, die kan het dan naar de markt brengen. We hoeven niet per sé zelfstandig te blijven. Onze toegevoegde waarde is het ontwikkelen van het medicijn. De farmabedrijven zijn beter toegerust om een product naar de markt te brengen.’
    
    Het medicijn van AM-Pharma heeft een ontstekingsremmende functie en voorkomt verdere celschade aan de nier. Het medicijn vangt potentieel slechte stoffen voor de nier weg en reduceert de ontstekingen waardoor het immuunsysteem niet op hol slaat. Van den Berg: ‘Met name die dubbelslag wekt het vertrouwen van de beleggers. Er bestaat bovendien nog geen product voor in de markt terwijl er een grote medische behoefte is. Er zijn nog drie andere bedrijven die een soortgelijk in ontwikkeling hebben, twee in de VS en één in Denemarken. De ontwikkeling van ons product is het meest vergevorderd.’
    
    AM-Pharma heeft een soortgelijk medicijn in ontwikkeling voor de behandeling van ontstekingen aan de darmen. Van den Berg: ‘Ook voor die ontwikkeling is binnen ‘een jaar of twee’ kapitaal nodig en daarna eveneens een farmapartner.’

    AMSTERDAM - Vanuit zijn Europese marktleiderschap in de opslag van stamcellen uit navelstrengbloed slaat Cryo-Save zijn vleugels steeds verder uit. Enkele maanden geleden werd een niche in de vooraanstaande Amerikaanse markt aangeboord. En met de opening gisteren van een nieuw laboratorium kunnen de nog bescheiden activiteiten in Zuid-Afrika volgens topman Arnoud van Tulder tot bloei komen.
    
    De opslag van stamcellen bij particuliere bedrijven komt in Nederland en de ons omringende landen moeizaam van de grond. Weliswaar druppelen de succesvolle transplantaties voor moeilijk behandelbare ziektes steeds meer binnen, maar medici zijn terughoudend zolang een daadwerkelijke doorbraak nog niet in zicht is. En dat zorgt er weer voor dat ouders niet snel geneigd zijn €2400 neer te tellen voor de verwerking en de opslag voor 25 jaar. Daar komt nog meer dan €300 bij indien ze tevens het navelstrengweefsel willen laten invriezen, waar inmiddels tweederde voor kiest.
    
    De hoop die Cryo-Save biedt, wordt in Zuid-Europa veel beter omarmd. En volgens Van Tulder zijn er volop mogelijkheden om van het nieuwe laboratorium in Zuid-Afrika een succes te maken. „Wij kunnen de bovenlaag van de bevolking via spreekuren in privéklinieken goed bereiken en merken dat er behoefte is aan de opslag van stamcellen. Ook al vanwege de versterking op het verkoopvlak door de samenwerking met een lokale speler, kan Zuid-Afrika volgend jaar uitgroeien tot ons vierde land qua volume.”
    
    Vanuit Zuid-Afrika wil Cryo-Save uitbreiden naar andere Afrikaanse landen. „We gaan verkoopkantoortjes opzetten, maar zien dit vooral als een ontwikkeling voor de langere termijn.” Ook voor de VS heeft het bedrijf er bewust voor gekozen niet overambitieus van start te gaan, zo maakt de topman duidelijk. „Wij willen hier niet direct de concurrentiestrijd aangaan met de top drie, omdat we dan vele miljoenen zouden moeten investeren. In plaats daarvan zijn we onlangs via een lokale partner gestart met ons vorig jaar in Zuid-Europa gelanceerde nieuwe product Cryo-Lip, om zo naamsbekendheid op te bouwen.”
    
    Cryo-Save biedt volwassenen hierbij de mogelijkheid om stamcellen uit vetweefsel op te slaan dat onder meer uit liposuctie wordt verkregen. „Onze ervaringen tot dusverre laten zien dat er vooral kansen liggen in toekomstig gebruik bij plastische chirurgie, waaronder borstreconstructie bij vrouwen en gezichtsverjonging. Wij zijn nu bezig met het opleiden van doktoren en denken in het vierde kwartaal de eerste klanten te krijgen, waarna de VS in dit segment snel groter kan worden dan in Europa”, aldus Van Tulder.
    
    Ook voor de al langer bestaande activiteiten ziet de topman volop mogelijkheden. „De marktomstandigheden in ons belangrijkste land Spanje zullen voorlopig moeilijk blijven. Wij denken echter marktaandeel van concurrenten te kunnen afsnoepen, mede door de versterking van onze samenwerking met een bedrijf dat bloedonderzoek voor ziekenhuizen verricht. Verder hebben de in het eerste halfjaar doorgevoerde veranderingen ervoor gezorgd dat India zich in juli en augustus goed herstelde. En de vestiging in Dubai betaalt de €1 miljoen die we twee jaar geleden moesten afboeken weer geleidelijk terug.”
